Te Restriction Fragment Length Polymorphism of Chromosomal DNA of Mycobacterium Tuberculosis Isolated in Korea




Abstract
Mycobacterium tuberculosis is a acid-fast, alcohol-fast, aerobic, nonspore forming and non motile bacillus. Recently, tuberculosis has taken a new turn in AIDS patients with a greater tendency toward more rapid progression. This study
investigated
the
value of IS6110-based DNA probe in epidemiology of tuberculosis. DNA fingerprinting assay with Mycobacterium tuberculsis has been a powerful epidemiologic tool. Such comparision will facilitate the investigation of the international transmission
of
tuberculosis and may identify specific strains with unique properties such as high infectivity, virulence, or drug resistance.
M. tuberculosis strains confirmed from BACTEC system were grown in 7H9 Middle brook medium supplemented with OADC(Difco) at 37¡Éfor 1 week. Chromosomal DNA was digested with restriction enzyme Pvu¥±. Southern blots were hybridized by using
digoxigenin-labeled IS 6110 as probe. IS 6110 restiction fragment length polymorphism(RFLP) analysis of various strains were performed.
Chromosomal DNAs of 68 different mycobacterial showed strain specific patterns by RFLP analysis. RFLP analysis of sixty-eigh M.tuberculosis strains showed bands with molecular sizes of 0.9 to 17.6-Kb range. RELP analysis of sixty-eight strains
showed
that each strain contained between 3 and 17 bands. RFLP analysis of sixty-eight strains showed 46 fragment. We may conclude that tuberculosis in Korea is not caused by one strain of M.tuberculosis and this RFLP methed could be useful for the
epidemiologic investigation in case of hospital infection and international comparison.
